Bioinformatic analysis and characteristics of glycoprotein C encoded by the newly identified UL44 gene of duck plague virus.

چکیده
Glycoprotein C is one of the duck plague virus (DPV) glycoproteins and is encoded by the DPV UL44 gene. DPV glycoprotein C (DPV-gC) comprises 431 amino acids with a putative molecular mass of 47.35 kDa. Sequence analysis indicated that the protein possesses typical characteristics of type-I membrane glycoproteins, containing an N-terminal signal sequence, an external domain, a C-terminal membrane anchor region, and a short cytoplasmic domain. Comparisons of 22 alphaherpesvirus-gC protein sequences revealed eight conservative Cys-residue sites, which may play a crucial role in the biological functions and structural stabilization of the DPV-gC protein. Estimates of potential antigenic epitopes and secondary structure identified four B cell dominant epitopes, which are located at amino acids 68-71, 87-91, 369-352, and 372-374. A model for the structure of DPV-gC was derived by associating its predicted secondary and three-dimensional structures. In conclusion, these results will provide a basis for further functional studies of DPV-gC, establishing novel clinical diagnoses of DPV, and in the development of a new DPV vaccine.

Molecular Farming, an Effective System for the Production of Immunogenic Crimean-Congo Hemorrhagic Fever Virus Glycoprotein
The main aim of this study was to obtain the Crimean-Congo hemorrhagic fever virus (CCHFV) glycoprotein, through either stable transgenic plants or using a transient expression system, and determine the yield, quality and finally the immunogenicity of the plant-made CCHFV glycoprotein in a mouse model. We designed and synthesized a codon-optimized G1/G2 gene from the G1 and G2 parts of the CCHF...
متن کاملDiagnosis and characterization of duck beak atrophy and dwarfism syndrome in Chongqing of China
Duck beak atrophy and dwarfism syndrome (BADS) is a newly emerged disease in China since 2015. In October 2017, an unidentified disease occurred in Cherry Valley ducks, Chongqing municipality, the southwest of China. The affected birds showed short beak and growth retardation clinical signs. The disease caused approximately 20.00% morbidity and serious weight loss due to retarded growth. In ord...

Background and Aims: The aim of this study was cloning and expression of rabies virus glycoprotein by a eukaryotic expression plasmid pcDNA3.1(+) in BSR cell line. This construct might be used for a potential DNA vaccine. Materials and Methods: Glycoprotein gene was synthesized and cloned into pBluescript vector and then sub cloned into eukaryotic expression vector (pcDNA3.1(+)). After verifica...
متن کاملConstruction of an Eukaryotic Expression Vector Encoding Herpes Simplex Virus Type 2 Glycoprotein D and In Vitro Expression of the Desired Protein
To construct of an eukaryotic expression vector encoding herpes simplex virus type 2 (HSV-2) glycoprotein D (gD2), an Iranian isolate of HSV-2 was propagated in HeLa cell line and its DNA was extracted and used as template in polymerase chain reactions (PCR), to amplify gD2 gene. Primers were designed and the restriction enzyme sites for EcoRI and XhoI were considered at their 5′ ends respectiv...
متن کاملA Sensitive Neutralization Assay for Influenza C Viruses Based on the Acetylesterase Activity HEF Glycoprotein
Influenza C virus possesses specific neuraminate-O-acetylesterase as a receptor-destroying function. This enzymatic activity of the viral glycoprotein HEF (Hemagglutinin, esterase activity and fusion factor) can be visualized in situ by the use of distinct color substrates. Hereby the localization, as well as the quantity of synthesized HEF protein is detectable. We further developed the estera...
